Cytotoxic Effect of L-Methioninase from Brevibacterium linens BL2 in Combination with Etoposide against Glioblastoma Cells

L-methioninase degrades methionine, which is essential in methionine-dependent cancer cells, resulting specific cell death. Normal cells can synthesize their own methionine amino acids even the absence of exogenous methionine. This selective targeting makes a promising therapeutic candidate for cancer. In this study, was partially purified from Brevibacterium linens BL2. The activity enzyme found as 3.055 units/mg. IC50 values (24 h) were 5.792 units/mL U87MG line and 5.215 T98G line. When etoposide used combination, synergistic cytotoxic migration inhibition effects on alongside decreased Mouse Embryonic Fibroblast HaCaT compared to alone observed. Additionally, colony numbers significantly reduced by administration after 21 days incubation. Furthermore, suppressed expression levels survivin c-Myc while increasing level Caspase-3 both glioblastoma lines. These enhanced when combination with etoposide. investigation reveals that not only exhibited but also anti-proliferative demonstrating fewer adverse normal cells.
